The tourists were blown away by the Springboks in the first Test, losing 38-16 in Durban.
There were 10 Lions in the starting line-up at Kings Park, but some of them are set to pay the price for a woeful performance with prop Adam Jones, scrum-half Mike Phillips and wing Alex Cuthbert among those in danger of the chop.
Youngsters who have impressed on tour, including Samson Lee, Gareth Davies, Matthew Morgan and Dan Baker, are all in contention for starting roles in Nelspruit.
Assistant coach Robin McBryde said: “Yes, there will be changes. Yes, some of them could be Lions players.
“It’s not a God given right that you’re given the opportunity and the honour to represent your country; that’s not something that we give away lightly.
“If you don’t produced the goods, then you don’t deserve to be on the field.
“We can’t shy away from making tough calls and tough decisions. You’ve got to put the team first and what’s best for the team.
“Selection will be made on the merit of what the players can bring to the table.”
